摘    要:参照B.J.Smirb(1984)介绍的蛋白质SDS—PAGE方法,调整了凝胶浓度、交联度及催化剂用量,提出了完善的样品制备程序,并研究了温度对凝胶配比及电泳时间的影响。在大量小麦整粒SDS—PAGE实验的基础上,又对半粒无胚籽粒电泳进行了细致的研究,摸荣出了一套适合于小麦整粒与半粒的高分子量(HMW)麦谷蛋白亚基SDS—PAGE方法。通过对5071份整粒种质资源和1600余份半粒杂交后代的分析,结果表明,该法成功率高,结果准确,具有操作简便、快速、分辨率好,制成干板不扩散易保存等优点。认为是一套适合我国仪器设备和药剂等实验条件的HMW谷蛋白电泳方法。是品质鉴定和后代品质筛选的可靠手段。

Abstract:A new SDS-PAGE method of high molecular weight (HMW) subunit of wheat glutenin was found, through reference to the method of protein SDS-PAGE by B. J. Smith (1984). The new method can analyse a full-single grain or a semisingle one. According to the electrophoretograms of 5071 full-single wheat varieties and more than 1600 semi-single hybrid progenies, it has been proved to have characters of high resolving power, appropriate gel strength, simple and rapid to use and bands in dry slab without spreading. Thus,it was regarded as a practical technique for separating HMW wheat glutenin protein, suitable for the home -made electrophoretic instrument and reagents.
