耐热丝氨酸蛋白酶改性大豆分离蛋白分散性条件研究

摘    要:利用耐热丝氨酸蛋白酶对大豆分离蛋白进行改性,提高其分散性。采用响应面实验设计,以加酶量、底物浓度、pH值和酶解温度为实验因素,以分散度指标为响应值,建立数学模型,优化酶解工艺参数。结果表明:最佳的酶解条件为:加酶量为4 070 U·g-1、底物浓度6.0%、反应温度71℃、pH 9.0,该条件下得到改性大豆分离蛋白的分散度为12.78,与未改性大豆分离蛋白分散度相比提高了2.09倍。

Abstract:The soy protein isolate was modified to improve dispersion of Heat-serne protease. The response surface was designed to experiment with enzyme dosage, substrate concentration, reaction temperature and pH as the factors and the dispersion index as response value, and establish mathematical model and optimize process parameters of enzymatic hydrolysis. The results indicated that the best hydrolysis conditions were enzyme dosage of 4 070 U ·g^-1,substrate concentration of 6.0%,reaction temperature of 71 ℃, pH 9.0.Under these conditions,the dispersity of soy protein isolate for modification reached 12.78,and compared to the dispersion of unmodified soy protein isolate improving 2.09 times.
