青钱柳愈伤组织培养条件研究的优化

摘    要:本试验研究了青钱柳愈伤组织培养中0.1%升汞溶液的最佳消毒时间、最佳外植体选择和最适基本培养基。结果表明:以叶片做为外植体,0.1%升汞溶液的消毒时间应掌握在3~4min之内,以茎段做外植体,消毒时间应掌握在14min左右。从愈伤组织诱导率上看,以当年生幼嫩茎段做外植体诱导效果更佳。MS和改良后的MS在特定的激素配比下均可诱导出健康的愈伤组织,以改良MS培养基+2.0mg/L6-BA+0.1mg/LNAA愈伤组织诱导率最高达83.33%,更适合青钱柳茎段的培养。

Abstract:The optimal sterilization time of 0.1% corrosive mercuric chloride, the optimal explant and the optimal basic culture medium were studied in callus culture of Cyclocarya paliurus Iljinsk. The study showed: the optimal sterilization time of 0. 1% corrosive mercuric chloride was 3 ~ 4 minute when stem section was explanted, while the optimal sterilization time was about 14 minute when the stem was used. The underyear young stern section was the most suitable explant in inductivity of calli. When we append specifically exogenous hormone to MS and reformative MS, robust calli were all induced, but the reformative MS+2. 0 mg/L 6--BA+0. 1 mg/L NAA was the best culture medium , inductivity of calli was 83.33%.
