继代培养中离体甜菜的倍性变化与增殖

摘    要:用卡宝品红法检测了不同来源离休甜菜的不同器官在不同年度间细胞焙性的变化。并统计分析了年度间变化的差异显著性及不同倍性优势细胞比率与增殖能力之间的相关性,结果表明,甜菜离休植株茎尖细胞倍性在继代培养中相对稳定,而根尖细胞倍性在年度间有显著变化。由未授粉胚珠培养再生的胚性细胞团,其细胞染色体容易自然加倍;再生的单倍体植株,其茎尖细胞为单倍体,而根尖细胞为二倍体。在所测年度内,离体甜菜细胞倍性的变化对其增殖能力无明显影响。通过茎尖和不断选择培养是离休甜菜保持遗传稳定性的有效手段。

Abstract:The variation on the cell ploidyof different organs from different plants in vitro were ex-amined with carfolfuchisn staning method during different years,and the correlation betweenthe difference significance of the variation and the multiplication of the plant were calcula-ted.The results bhow that in the subculture,the cell ploidy in the stem tip of plants in vitrocould remain relatively stable and the changes of cell ploidy in root tip of plant in vitro wassignificanl.In the embryonic cell clones and the haploid plants regenerated from unpollinatedovules,the chromosomal doubling of the former could occoured naturally and the cells in thestem tip of the latter were haploid but the cells in the root tip were diploid.There was no sig-nificant influence on the variation of cell ploidy being examined during different years for themultiplication of sugar beet in vitro. It is an effective method to ramain genetic stability ofsugarbel3t in vitro in subculture by the culture of the stem tip and belection constant at thetransference of plant in vitro.
