立达霉在草鱼体内的代谢机制研究

摘    要:[目的]研究立达霉在草鱼体内的代谢机制。[方法]采用高效液相色谱-串联质谱(HPLC-MS/MS)检测方法,检测草鱼在立达霉单次口灌200 mg/kg,浸泡9 mg/L剂量条件下,其肝脏组织中立达霉原药含量及代谢产物的种类,研究立达霉在草鱼肝脏组织中的代谢机制。[结果]在口灌组及浸泡组,肝脏组织中的立达霉以原药(M0)为主,高于75.00%;立达霉在草鱼体内代谢产物主要有4种,分别为代谢物N-(2-羧基-6-苯甲基)-N-(甲氧乙酰基)丙氨酸、N-[(2-羟甲基)-6-甲苯基]-N-(羟乙酰基)丙氨酸、N-(2,6-二甲苯基)-N-(羟乙酰基)丙氨酸和N-(2-羟甲基-6-苯甲基)-N-(甲氧乙酰基)丙氨酸甲酯,各代谢产物所占比例均小于10.00%。[结论]立达霉在草鱼肝脏组织中残留应以立达霉原药作为残留检测标识物。

Metabolic Mechanism Study on Ridomil in Grass Crap
Abstract:[Objective]To study metabolic mechanism on ridomil in grass crap.[Method]Using high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS),under the condition of single oral infusion of 200 mg/kg and soaked in 9 mg/L,detected the content of pro-totypic drug,kind of its metabolite in liver tissue and ridomil metabolic mechanism in liver tissue of grass carp.[Result]Both oral perfusion group and soaking group,ridomil in the liver tissue chiefly on prototype drug was highter than 75.00%.It's metabolites in liver mainly had four kinds, N-(2,6-dimethylphenyl)-N-(hydroxyacetyl)alanine,N-(2-hydroxymethyl-6-methylphenyl)-N-(methoxyacetyl)alanine methyl ester,N-(2-carboxy-6-methylphenyl)-N-(methoxyacetyl)alanine,N-[(2-hydroxymenthyl)-6-methylphenyl]-N-(hydroxyacetyl)alanine,respectively.The proportion of each metabolite was less than 10.00%.[Conclusion]Residual of ridomil in liver of grass carp should be based on prototypic drug as residue detection marker.
