盾壳霉孢子苹果渣培养研究

摘    要:以苹果渣为培养基,采用单因素试验研究苹果渣预处理方法、盾壳霉孢子种龄、初始pH、碳酸钙添加量和脲素添加量及其添加方式等对盾壳霉孢子生长的影响,优化培养条件。结果表明,将苹果渣用4倍水清洗1次,灭菌后接入3日龄液体种子,以及调节苹果渣pH为6.5,添加15 g.kg-1碳酸钙或25 g.kg-1脲素均有利于盾壳霉孢子的产生,每克培养基最多可产孢子1.1×109~1.2×109个。

Abstract:Cultivation conditions of Coniothyrium minitans spores in apple pomace were studied by single factor experiments,such as the pomace pretreatment,ages of Coniothyrium minitans spores,initial pH,the added amount and method of calcium carbonate and urea.The results showed that flushing apple pomace with 4 times of water for one time,using three-day-old seeds of PDB culture solution of Coniothyrium minitans,adjusting pH to 6.5,adding 15 g·kg-1 CaCO3 or 25 g·kg-1 urea are all helpful to the production of spores.The highest yield could reach to 1.1×109-1.2×109 spores per gram of culture medium.
