紫晶蜡蘑的营养优化及生长模型

摘    要:选出紫晶蜡蘑菌株(Laccaria amethystea-0704;简称L.a.-0704)适宜培养基,在L.a.-0704碳源及氮源营养选择的基础上,采用通用旋转回归设计进行培养基配方优化。得到L.a.-0704菌丝体培养的最佳配方为麦芽浸粉4.1 g/L,尿素1 g/L,蔗糖4.3 g/L,酵母浸膏3.5 g/L,CaCl20.05 g/L,(NH4)2HPO40.25 g/L,NaCl 0.025 g/L,MgSO4.7H2O 0.15 g/L,KH2PO40.5 g/L,FeCl3(w=1%)2 mL/L,VB1100μg/L,水1 000 mL,琼脂粉12 g/L。L.a.-0704营养利用范围广,不仅能利用单糖、低聚糖和无机氮等速效碳、氮源,而且能利用多糖、多元醇和有机氮等长效碳、氮源,L.a.菌丝生长符合Loginstic模型,其菌丝生长最适碳、氮源分别为麦芽浸粉和尿素。

Abstract:The objective of the study was to optimize the medium for culturing strain "L.a.-0704" of Laccaria amethystea The medium was optimized based on the culturing property studies on different media,and the suitable C and N were studied in detail thereafter.The results showed that the hyphae of strain ’0704’ could obtain nutrition from wide range sources including rapidly available C or N,such as monosaccharide,oligosaccharide,or long-acting C or N,such as polysaccharide,polylol,and organonitrogen.The hypha growth met the Loginstic pattern.The appropriate C and N are malt extract powder and urea.The optimum medium for the hypha growth is malt extract powder 4.1 g/L,and urea 1 g/L,sucrose 3 g/L,Yeast Extract 3.5 g/L,CaCl2 0.05 g/L,(NH4)2HPO4 0.25 g/L,NaCl 0.025 g/L,MgSO4·7H2O 0.15 g/L,KH2PO4 0.5 g/L,FeCl3(w=1%) 2 mL/L,VB1 100 μg/L,water 1 000 mL,agar 12 g/L,Strain-0704 of Laccaria amethystea(Bull.ex Gray) Murr.digested not only available nitrogen and carbon,such as monosaccharide,oligosaccharides,inorganic nitrogen,but macromolecular compound,such as polysaccharide,polyol and organic nitrogen.The growth of L.amethystea hypha was in accordance with Loginstic model.The appropriate nitrogen or carbon was urea or malt powder respectively.
