中度嗜盐菌Nesterenkonia sp.DF-1产淀粉酶发酵条件优化的研究

摘    要:[目的]摸索出Nesterenkoniasp.DF-1菌种的产淀粉酶的优化发酵条件,为发酵工业中淀粉酶的大规模生产提供理论依据。[方法]采用单因素和多因素正交试验的方法,考察发酵时间、温度、初始pH、盐度、碳源和氮源的改变对发酵产淀粉酶的影响。[结果]在装液量100ml/250ml条件下,得到产淀粉酶最佳条件为:玉米粉1%,酵母膏2%,初始pH8.O、NaCl浓度3%、37.5℃、160r/min振荡培养72h。[结论]该研究找到了产淀粉酶的最佳发酵条件。

Abstract:[ Objective] It was necessary for Nesterenkonia sp. DF-1 to optimize the fermentation conditions for production of amylase in order to provide theoretical basis for the mass production of amylase in fermentation industry. [ Method] The single factor method and multi-factor orhogonal experiment were adopted to investigate the effect of a set of variables, including fermentation time, temperature, initial pH, salinity, the change of carbon source and nitrogen source upon the influence of fermentation to produce amylase. [ Result] When liquid medium volume was 100 ml/250 ml, the optimal fermentation conditions were corn meal 1% , yeast extraction 2% , initial pH 8.0, NaCl concentration 3% , temperature 37.5 ℃ and shaking culture for 72 h at 160 r/min. [ Conclusion] The best fermentation conditions for Nesterenkonia sp. DF-1 to produce amylase were obtained.
