摘    要:本研究以云南野生金丝梅(Hypericum patulum)带休眠芽的茎段为外植体,研究了不同植物生长调节剂对其离体培养的影响,以建立金丝梅离体快繁技术体系。研究结果表明,腋芽诱导的适宜培养基为MS+6-BA 2.0 mg/L+NAA 0.1 mg/L,诱导率可达90%;适合腋芽增殖的培养基为MS+6-BA 1.0 mg/L+NAA 0.1 mg/L+GA30.1 mg/L,增殖系数可达5.12;最佳生根培养基为1/2MS+IBA 0.5 mg/L+活性炭0.1 g/L,生根率为96.69%。炼苗移栽到草炭土∶珍珠岩=2∶1的基质中,成活率达100%,移栽到苗圃后成活率为100%。研究结果为金丝梅高效快速繁殖以及优良品种选育提供科学依据。

Abstract:In this study,the stem segments of Hypericum patulum with axillary buds which were collecting from Yunnan province were used as explants to study the effects of different plant growth regulators on their culture in vitro,and to establish the tissue culture and rapid propagation system of Hypericum patulum.The results showed that the optimal medium for germination of stem with axillary buds was MS+6-BA 2.0 mg/L+NAA 0.1 mg/L,with a 90%germination rate;the optimal medium for shoot multiplication was MS+6-BA 1.0 mg/L+NAA 0.1 mg/L+GA30.1 mg/L,and the multiplication coefficient could reach to 5.12;the optimal rooting medium was 1/2 MS+IBA0.5 mg/L,with a 96.69%rooting rate.After the tissue cultured,the seedlings were transplanted to the matrix composed of turfy soil and perlite(2:1),and the survival rate was up to 100%.Then,the plants were transplanted to nursery,and the survival rate was up to 100%.This research could provide scientific basis for the efficient and rapid reproduction of Hypericum patulum and the selection and breeding of fine varieties.
