摘    要:以务川草石蚕茎尖为外植体,研究了茎尖长度、热处理温度和时间对草石蚕脱毒的影响,并建立草石蚕脱毒快繁技术体系。结果表明,随草石蚕茎尖长度减小,脱毒率升高,但成苗率降低;以0.5 mm茎尖长度为最佳,成苗率和脱毒率分别为51.7%和38.7%。0.5 mm茎尖经50℃处理30 min后,其脱毒率可提高到60.0%。1.0 mm茎尖在40℃培养20 d后的脱毒效果最佳,可达88.37%。已脱毒芽增殖的最佳培养基为MS+NAA 0.3 mg/L+6-BA 5 mg/L+GA3 1.5 mg/L;生根最优培养基为1/2MS+NAA 0.2 mg/L+GA3 0.5 mg/L。本研究建立了草石蚕脱毒快繁的技术体系,可为草石蚕脱毒苗的快速繁殖提供技术参考。

Abstract:The effects of bud length,thermotherapy temperature and time on the virus elimination and rapid propagation system were investigated in Stachys sieboldii in the present study.The results showed that the percentage of virus elimination rose,and the seedling percentage fell with the bud length decreasing.0.5 mm bud length was the best,and the seedling percentage and virus elimination percentage were 51.7%and 38.7%,respectively.The virus elimination percentage would increase to 60.0% when the 0.5 mm buds were treated under 50℃ for 30 minutes.The virus elimination was best when the 1.0 mm buds were treated under 40℃ for 20 days,of which the percentage reached 88.37%.The best proliferation medium for virus-free adventitious buds was MS+NAA 0.3 mg/L+6-BA 5 mg/L+GA31.5 mg/L.The most suitable medium for rooting was 1/2 MS+NAA 0.2 mg/L+GA30.5 mg/L.This experiment established a technical syste m for virus elimination and rapid propagation in Stachys sieboldii,which could provide technical reference for the rapid propagation of virus-free seedlings.
