辣椒YABBY基因家族的全基因组分析

摘    要:YABBY基因家族成员在叶和子叶,花器官和胚珠珠被的发育中起关键作用。拟南芥中,YABBY基因家族已经有较透彻的研究。本研究以拟南芥YABBY基因为检索序列,挖掘辣椒‘遵辣一号’和其野生祖先‘Chiltepin’基因组中的YABBY基因序列,并进行染色体定位、系统进化及可能的功能等分析。分析结果表明,辣椒基因组中含有7~8个YABBY基因,和墨西哥野生祖先‘Chiltepin’相比,栽培品种‘遵辣一号’YABBY基因大体保持了一致,但也有重要的区别。‘Chiltepin’九号染色体上的基因Capang09g000047,与‘遵辣一号’中一号染色体上的Capana01g003578序列相近,位于同一分支。可能是由于‘Chiltepin’九号染色体上的基因Capang09g000047发生了移位产生。此外,‘遵辣一号’中十一号染色体上的Capana11g002002,在‘Chiltepin’中没有对应基因。这一基因与‘遵辣一号’和‘Chiltepin’一号染色体上的同源基因序列非常相似。对YABBY基因家族在‘遵辣一号’开花期植株的幼叶、成熟叶片、花芽和成熟花中的表达分析,发现YABBY基因家族成员在叶和花中广泛表达,不同成员间表达量具有较大差异。本研究为辣椒基因组中YABBY基因家族的功能研究提供了理论基础。

Abstract:Plant specific YABBY gene family plays a key role in the development of leaves and cotyledons,floral organs and ovules.Arabidopsis thaliana YABBY gene family has been thoroughly studied.In this study,the YABBY genes of the Arabidopsis thaliana were used to identify the YABBY genes in'Chiltepin'genome,and analyze their chromosomal loci,phylogenetic and possible function.The genome of pepper contains from 7 to 8 YABBY genes.Compared with the wild ancestor,'Chiltepin'in Mexico,the cultivar'Zunla-1'YABBY gene were generally conserved,but there were also important differences.The gene Capang09g000047 on'Chiltepin'chromosome 9 is similar to the Capana01g003578 sequence on'Zunla-1'chromosome 1.In addition,Capana11g002002 on'Zunla-1'chromosome 11 has no corresponding gene in'Chiltepin'.This gene is very similar to the homologous genes on the chromosome 1.The expression analysis of the YABBY gene family in the young leaves,mature leaves,flower buds and mature flowers of the'Zunla-1'plants at flowering stage revealed that the YABBY gene family members were widely expressed in leaves and flowers.The expression levels between different members have large differences.This study provided theoretical foundation for the functional study of the YABBY gene family in the genome of pepper.
