培养条件及贮藏方式对郁香忍冬离体花粉萌发的影响

花粉是植物有性生殖遗传物质传递的载体,其活力直接影响遗传物质传递的效率,花粉正常萌发是保证受精作用顺利进行的前提,人工模拟柱头中的花粉萌发环境,是检测花粉活性最可靠的方法。本研究以郁香忍冬离体花粉为试验材料,研究了不同蔗糖浓度(15%~45%)、硼酸浓度(100~300 mg/L)、不同培养温度(9℃,12℃,15℃,18℃)及贮藏方法对花粉萌发率的影响。结果表明:郁香忍冬花粉在30%蔗糖+100 mg/L硼酸溶液中,12℃条件下培养,萌发率最高。花粉短期贮藏的最佳方式为冷冻(-20℃)贮藏,冷藏(5℃)次之,而室温条件不适宜郁香忍冬花粉贮藏。该结果可为忍冬属植物人工杂交育种工作的开展提供参考依据和技术支撑。

Effect of Cultural Condition and Storage Method on the Pollen Germination in Vitro of Lonicera fragrantissima

Pollen is the carrier of genetic material transfer in sexual reproduction of plants,and its vitality directly affects the efficiency of genetic material transfer.The pollen germinated normally is the premise for successful implementation of the fertilization.Artificial simulation of pollen germination environment in stigma is the most reliable method to detect pollen activity.In order to screen the optimum culture conditions for pollen germination in vitro of Lonicera fragrantissima,and to study the effect of different storage methods on pollen activity.In this study,the pollen of Lonicera fragrantissima was used as experimental material,and the effects of different sucrose concentration(15%~45%),boric acid concentration(100~300 mg/L),different culture temperature(9℃,12℃,15℃,18℃)and storage methods on pollen germination rate were studied.The results showed that the pollen germination rate are the highest on the medium supplement with 30%sucrose+100 mg/L boric acid,culturing at 12℃.The best method for short-term storage of pollen is cryopreservation(-20℃),followed by refrigeration(5℃),but the room temperature was not suitable for the storage of the L.fragrantissima pollen.The results of this study can provide reference and technical support for artificial cross breeding of Lonicera.
