摘    要:为研究ABA、KNO3和NH4NO3两种氮源比例及凝固剂对白刺花体细胞胚胎发育、成熟及萌发的影响。本研究利用白刺花下胚轴诱导的增殖良好的胚性愈伤组织为材料,通过ABA、氮源种类及比例和凝固剂对其体胚发生、成熟的对比试验研究,筛选最优的白刺花体细胞胚胎发育、成熟和萌发的培养基。结果表明:白刺花下胚轴诱导的愈伤组织经增殖培养得到增殖率为170%的胚性愈伤组织;添加ABA 15.0 mg/L的培养基上可使体细胞胚发生率达到85.21%,体细胞胚产生数量达112个/g,总胚数与对照相比为486.96%;氮源比例中,KNO3增加1倍,NH4NO3减半的培养基上体细胞胚发生率最高,达92.33%,体细胞胚产生数量最多,为139个/g,形成的体胚数是对照的124.11%;以琼脂和植物凝胶作为凝固剂的试验中,琼脂7.0 mg/L和植物凝胶为3.0 mg/L时,体细胞胚发育良好,从畸形胚及子叶胚的多少和愈伤褐化综合考虑,植物凝胶比琼脂好。不同发育期的体细胞胚萌发培养时,鱼雷胚转接最为合适,体胚萌发能力最高,为91.55%;体胚苗壮苗培养可使植株转化率高达93%,植株移栽成活率为92.92%。本研究可为白刺花体细胞胚胎发生技术和快速成苗体系提供基础资料。

Abstract:In order to study ABA,KNO3 and NH4NO3 type and ratio,coagulant that have impacts on development,maturation and germination of somatic embryo of Sophora davidii.This research used embryonic callus as materials from hypocotyl.The development,maturation and germination of somatic embryo was determined by comparative research of different ABA,KNO3 and NH4NO3 type and ratio,coagulant.The optimum medium for somatic embryo development,maturation and germination and plantlet regeneration were screened.The results showed that the callus of the hypocotyl induced by the hypocotyl of Sophora davidii was cultured to obtain the embryogenic callus with the yield of 170%.Somatic embryogenesis rate reached 85.21%,and number of somatic embryos reached 112/g,compared with the control group,total number of embryos was 486.96%,higher than other treatments onMS medium added with ABA 15.0 mg/L.Regarding the nitrogen ratio,somatic embryogenesis rate reached 92.33%,and number of somatic embryos reached 139/g,compared with the control group,total number of embryos was 124.11%,higher than other treatments on MS medium added one fold KNO3 and half NH4NO3.Regarding the solidification agent as agar and plant gel,somatic embryos developed well on MS medium added 7.0 mg/L agar or 3.0 mg/L plant gel.Considering the number of abnormal embryos and cotyledon embryos and browning callus,plant gel was better than agar.Torpedo embryo was the most appropriate at different developmental stages of somatic embryo,which the rate of somatic embryo germination was up to 91.55%.In the culture of somatic embryo seedling,the plant conversion rate was 93%.Up to 92.92%of the transplanted plantlets were successfully survived one months in soil.This study provides a technical support technique for large-scale propagation of Sophora davidii embryo seedling.
