无角牦牛crtc3基因多态性及其与生长性状的相关分析

摘    要:为探究crtc3基因在无角牦牛群体中的遗传多态性,开展其与生长性状间的关联性。利用PCR-SSCP技术结合DNA测序对354头无角牦牛crtc3基因进行多态性分析,并运用SPSS 20.0软件对各基因型间不同月龄(6,12和18月龄)的体质量、体高、体斜长和胸围等生长性状指标进行关联分析。结果表明,在牦牛crtc3基因的第14内含子的第86041处发生C→T单个碱基突变,结合PCR-SSCP结果分析后,分为CC、CT和TT 3种基因型。关联分析发现,在6月龄,CC基因型个体体高显著高于TT基因型个体,CC基因型个体胸围显著高于CT基因型个体;在12月龄,CC基因型和CT基因型个体体高极显著高于TT基因型个体,CC基因型个体胸围极显著高于TT基因型个体;在18月龄,CC基因型个体体质量和体高显著高于TT基因型个体。综上所述,在牦牛crtc3基因上检测到第14内含子的第86041处发生C→T单个碱基突变,该突变位点对无角牦牛各时期的体质量、体高和胸围等生长性状等有显著影响,可以作为一种遗传标记用于辅助选择。

Abstract:To explore crtc3 gene polymorphism in the polled yak groups,its correlation with growth traits was carried out.Crtc3 gene polymorphism of 354 polled yak was analyzed using PCR-SSCP technology combined with DNA sequencing,and the relationship of growth character indexes,such as body weight,body length and chest circumference of different months(6,12 and 18 months)between genotype,was analyzed by the SPSS 20.0 software.The results showed that the C-T single base mutation occurred at the position of 86041 of the 14 th intron of crtc3 gene in yak,which was divided into CC,CT and TT genotypes combining the PCR-SSCP analysis.Association analysis showed that at the 6 months age,the body height of CC genotype individuals was significantly higher than that of TT genotype individuals,and the chest circumference of CC genotype individuals was significantly higher than that of CT genotype individuals.At the 12 months age,the body height of CC genotype and CT genotype individuals was significantly higher than that of TT genotype individuals,and the chest circumference of CC genotype individuals was significantly higher than that of TT genotype individuals.At the 18 months age,the weight and body height of CC genotypes were significantly higher than those of TT genotypes.In conclusion,the C-T single base mutation was detected at 86041 of the 14 th intron of crtc3 gene in yak,which had a significant impact on the growth characteristics of polled yaks at different stages,such as weight,body height and chest circumference,and could be used as a genetic marker of assisted selection.
