107速生杨遗传转化中最适培养基的筛选

摘    要:以107速生杨为试材,以氯化钠(NaCl)和卡那霉素(Kan)作为筛选指标进行遗传转化中最适培养基的筛选研究。对比转基因材料和对照材料的分化率和长势,并对转基因材料进行PCR检测,最终确定最适于遗传转化筛选的培养基。结果为氯化钠(NaCl)不能用作筛选指标,卡那霉素是最适筛选指标,我们得到的最适筛选培养基是加入100mg/L卡那霉素的分化培养基。

Abstract:Different concentrations of NaCl and Kanamycin were used for studying on selection of transgenic Poplar plantlets. Through comparing the differentiation rate and growth potential of transgenic plants and control, then transgenic plant proved by PCR analysis, we acquired optimal selective medium in our transformation system. NaCl can't be used for selective marker. Kanamycin can be used for selective marker in our system. And the optimal selective medium was modified MS with BA 0.5 mg/L, NAA 0.1 mg/L, ceftriaxone sodium 200 mg/L and Kanamycin 100 mg/L.
