摘    要:目的]探讨IGFBP-5基因在山羊出生后不同组织的发育性表达模式。方法]采用荧光定量PCR,分析了出生后0、15、30、60、90和120d36只南江黄羊(公、母羔各18只)的心脏、肝脏、脾、肺、背最长肌、半膜肌、臂三头肌和股二头肌样品中IGFBP-5mRNA水平。结果]母羔的IGFBP-5mRNA表达丰度显著高于公羔。各组织IGFBP-5mRNA表达量呈现肝脏〉肺〉脾(心脏)〉肌肉的趋势(P〈0.01),脾与心脏间差异不显著(P〉0.01),肌肉的表达值最低。随着日龄的增加,各组织中IGFBP-5mRNA丰度均呈现下降-上调-下降的发育性表达模式,而肝脏和肺中的表达模式具有明显的性别差异。结论]IGFBP-5mRNA主要在内脏,尤其是肝脏中合成,但在各组织的发育性表达模式一致,性别是一个重要的影响因素。

Developmental Expression Patterns of IGFBP-5 mRNA in Different Tissues of Goat

Abstract:Objective]The research aimed to investigate the developmental expression patterns of insulin-like growth factor-binding protein 5(IGFBP-5) gene in different tissues of postnatal goat.Method]The heart,liver,spleen,lung,longissimus dorsi,semimembranosus,arm triceps,and biceps femoris samples were collected from 36(18 male and female,respectively) Nanjiang Mongolian Grazelle at age of 0,15,30,60,90,and 120 days after birth.And the ontogeny mRNA levels of IGFBP-5 was analyzed by real time fluorescence quantitative RTPCR.Result]IGFBP-5 mRNA level in female goat was significantly higher than male goat,and the abundance of IGFBP-5 mRNA decreased dramatically in order of liver,lung,spleen or heart,and muscles.There was no significant difference between spleen and heart,and the abundance was lowest in muscles.IGFBP-5 mRNA in all tissues showed the variation tendency of down-up-down patterns with age,and the expression patterns had the obvious gender difference between liver and lung.Conclusion]The internal organs especially the liver are the main production sites of IGFBP-5 mRNA,while the developmental patterns are similar within these eight tissues,and gender plays an important role in abundance of IGFBP-5 mRNA.
