摘    要:肌细胞生成素(myogenin, MyoG)是生肌调节因子MRFs (MyoD、MyoG、Myf5和MRF4)家族的一员,它的表达具有控制成肌细胞融合的起始,促使成肌细胞增殖的作用,使单核成肌细胞转变成为多核的肌纤维,在MyoD家族中具有中心地位.本试验克隆了鹅MyoG外显子1序列,得到其片段长度为438 bp,与鸡MyoG基因的同源性为91.9%,与人、牛、猪等哺乳动物同源性为68%~70%,其氨基酸序列与鸡的同源性达到93.8%.核酸进化树显示,鹅、鸡较早地与哺乳类动物分化开来.分析了该区域编码氨基酸的亲水性、相应区域位于表面可能性和平均电荷,以及密码子的使用策略.

Abstract:Myogenin gene(MyoG) is one of myogenic regulatory factors(MRFs) such as MyoD,MyoG,Myf5 and MRF4.It can control the start of myoblasts fusion,and induce the myoblasts proliferation,and then turn the monocaryon myoblasts to the multinuclear myotubes,thus MyoG has important function in MRFs.The total DNA from serum of goose was extracted,and a pairs of primer named P_1,P_2 was designed to amplify the MyoG gene according the MyoG gene sequence of chicken in GenBank.As a result a 438 bp sequence of MyoG gene was amplified,cloned and sequenced.Also we analyzed the sequence between chicken,homo,pig,mouse,cattle and dog,it showed there was 91.9% homology at the nucleotide level between goose and chicken which is the highest among goose and other species.The homology between goose and mammals(human,bovine and pig,et al) is between 68% to 70%.And there was 93.8% homology at the amino acid level between goose and chicken.The phylogenetic tree shows that goose and chicken differentiated from mammals earlier.Aslo we analysed the hydrophilicity,surface probability,average charge and condon usage of amino acids coded by exon 1 of MyoG gene in goose.
