首个核果类果树病毒RT-PCR检测试剂盒的研制与应用

摘    要:以带毒(阳性)和非带毒(阴性)的GF305为试材,利用已建立的总RNA提取方法提取试材中的总RNA,根据3种病毒(ACLSV、PNRSV和PDV)外壳蛋白基因序列设计寡核苷酸引物,采用RT-PCR技术研制3种病毒检测试剂盒,以达到最优化检测.并应用该试剂盒对果园中存在的病毒进行检测.结果显示,该试剂盒检测病毒速度快(6~7h)、灵敏度高(较ELISA法高8 000倍)、专一性强、检测成本较低,是一种有推广价值的核果类果树病毒检测方法.该试剂盒已申报了国家发明专利,初步审查合格.

Abstract:The GF305 was chosen as experimental material. The total RNA was extracted by the method established. The specific DNA primers were designed based on the coat protein gene sequences of ACLSV, PNRSV and PDV, and the first virus detection Kit by RT-PCR in stone fruits was prepared and applied to detected these trees of cherry, peach and apricot planted in the field. The results showed that the kit was more sensitive and rapid than ELISA, and it had a higher specificity and lower cost accouting.
