摘    要:为改良拮抗姜瘟病菌的芽孢杆菌,从未发病土壤分离得到1株对茄科雷尔氏菌(Ralstonia solanacearum)等8种病原菌具有较强拮抗能力的蜡状芽孢杆菌菌株L1.为进一步改进该菌的拮抗活性,采用紫外线(UV)、甲基磺酸乙酯(EMS)、吖啶橙等诱变因素对L1菌株进行诱变改良.结果表明:改良后菌株的拮抗性能显著提高,其中菌株EMS-2和UL-1的拮抗蛋白浓度分别比L1菌株提高30.38%和29.18%,其差异均达极显著水平;L1对UV照射十分敏感,尤其在前60 s,而90 s的照射处理正突变率最高,达24.8%;吖啶橙处理L1后,没有筛选到发生正突变的菌株;L1在EMS处理80 min后,致死率达到90%左右,因此,为大量获取正突变菌株,应采用较低剂量的EMS(0.16 mol?mL-1)、长时间处理程序(60 min为宜).

Abstract:To improve the resistance of Bacillus cereus to Ralstonia solanacearum,an antagonistic bacterium belonging to B.cereus,named as L1,was isolated from the ginger-growing soil,which was resistance to R.solanacearum at some degree.Ultraviolet rays(UV),ethyl methane sulfonate(EMS) and acridine orange (AO) were used as mutagens to treat strain L1,and then the mutants were identified and screened.The results showed that the antagonistic abilities of mutant strains were significantly enhanced compared with the orig...
