摘    要:采用高效液相色谱法和苯酚硫酸法分别检测北冬虫夏草和冬虫夏草中虫草素和虫草多糖含量。HPLC色谱条件为:色谱柱为Waters NOVA—PAK C18(3,9mm×300mm,4μm);流动相为水:甲醇=90:10;流速1.00mL·min^-1,检测波长260nm;虫草多糖检测条件为:在待测样品中加入硫酸10mL,显色1min后,在酶标仪上用490nm波长测定样品的吸光值。结果表明:高效液相色谱法的检测虫草素的回归方程为Y=-7.066×10^6+3.206×10^6X,R=0.9999(n=3),虫草素的平均含量为北冬虫夏草子实体1.137%,菌丝体0,7162%.冬虫夏草中0.000523%,苯酚硫酸法检测虫草多糖的回归方程为Y=0.09889+1.161X,R=0.9964(n=7),虫草多糖在不同材料中的含量分别为北虫草子实体3.35%,菌丝体3.05%,冬虫夏草7.83%。该研究为进一步研究冬虫夏草替代品奠定了基础。

Abstract:The contents of cordycepin and Cordyceps polysaccharide (CP) in Cordyceps militaris and Cordyceps sinensis were detected by high performance liquid chromatography (HPLC) method and phenol-sulfuric acid method. The chromatography conditions for HPLC method were as follows:the column was Waters NOVA-PAK C18 (3.9 mm × 300mm, 4 μm), and mobile phase was composed of Water (A) and Methanol (B) (90% A, 10% B) with flow rate of 1.0 mL/min and 260 nm detection wavelength. The phenol-sulfuric acid method was used with an extracting condition which was as follows: the color time was lmin with 10 mL sulfuric acid added to the sample, then the sample was measured at 490nm wavelength. The detecting results showed that the regress equation of HPLC detection was Y=-7.066× 10^6+3.206×10^6X, R=0.9999 (n=3) and the average cordycepin content in the fruit-bodies of Cordyceps militaris was 1.137%, while in mycelia of Cordyceps militaris 0.7162% and 0.000523% in Cordyceps sinensis. The regress equation of phenol-sulfuric acid method for detecting CP was Y=0.09889+1.161X,R=O.9964(n=7). And CP contents of Cordyceps sinensis was 7.83%, in fruit-bodies of Cordyceps militaris was 3.35% and in mycelia of Cordyceps militaris was 3.05%. The work demonstrated here establishes foundation for further research on Cordyceps substitute.
