摘    要:[目的]建立HPLC测定马比木根中喜树碱含量的方法。[方法]HPLC法测定,色谱柱为Diamonsil C18柱(5μm,250mm×4.6mm),流动相为乙腈∶水(30∶70),检测波长254nm,流速1.0ml/min,柱温为室温。[结果]喜树碱含量在0.046~0.230mg/ml范围内线性关系良好;仪器精密度良好,其RSD值为1.30%;试验的重现性良好,其RSD值为0.87%;试验的稳定性良好,其RSD值为2.30%,说明喜树碱在48h内比较稳定。加样回收率试验,重复5次,计算得到平均回收率为99.2%,RSD值为1.20%。湖北恩施和重庆黔江产马比木根中的喜树碱含量分别为0.1436%和0.1448%。[结论]该方法分离效果好、专属性强、分析速度快、灵敏度高,可作为马比木根中喜树碱含量测定的方法。

Abstract:[Objective]The aim was to establish a HPLC method for determining the camptothecin in roots of Nothapodytes pittosporoides. [Method]Diamonsil C 18 column (250 mm×4.6 mm,5 μm) was used,the mobile phase was consisted of CH3CN-H2O (30∶70),the UV detector wavelength was 254 nm,and flow rate was 1.0 ml/min. [Result]The calibration curves showed that a good linearity has been in the range of 0.046-0.230 mg/ml with RSD as 1.30%; the reproducibility of the test was good,with RSD as 0.87%; and the stability was also good,with RSD as 2.30%,which showed that the camptothecine is stability within 48 h. The average recovery experiment,repeated for five times,calculated the average recovery was 99.2%,RSD value was 1.20%. Camptothecine contents were 0.143 6% in Enshi sample and 0.144 8% in Qianjiang sample. [Conclusion]The method for determining the camptothecine in roots of N. pittosporoides was rapid,sensitive and accurate.
