高活性魔芋葡甘聚糖酶产生菌B23的鉴定及培养条件优化

摘    要:葡甘聚糖酶产生菌B23分离自哺乳动物的内脏,经鉴定可能为芽孢杆菌属一新种。该菌生长的最适pH为中性环境,可耐受60℃高温,且对NaCl有很好的耐受力。摇瓶培养时最佳氮源为蛋白胨,魔芋葡甘聚糖做碳源诱导酶的产生,0.2%K2HPO4有利于B23的生长及提高产酶的活性。通过正交试验优化的培养条件为温度40℃、初始pH值7.0~7.2、装液量20%、接种量10%。这些数据为扩大生产葡甘聚糖酶提供了参考。

Abstract:The bacterium producing glulcomannanase named B23 was screened from mammal viscera. By our identification, it was possibly a new species of Bacillus. This bacterium had the optimum pH of neutrality and excellent resistance to heat(60) and hypersaline(10% NaCl). The optimum nitrogen source of rotating culture medium was peptone and carbon source was KGM which could induced the product of glucomannanase. 0.2% K2HPO4 could increase the biomass and the activity of glucomannanase. According to the orthogonal experiment results, The optimized culture condition was temperature 40, initial pH 7.07.2, medium volume 20% and seed culture 10%. The results could provide the glucomannanase production in large scale with references.
