Establishment and Application of Fluorescence Quantitative PCR Detection Method for Apis mellifera and Apis cerana Honey

Honey is mainly produced by Apis mellifera and Apis cerana in China.The price of A.cerana honey is usually three to five times higher than that of A.mellifera honey due to limited productivity and good nutritional value and health function.Some dishonest enterprises and beekeepers mislabeled A.mellifera honey as A.cerana honey,or incorporated A.mellifera honey into A.cerana honey.Therefore,it is necessary to distinguish between A.cerana honey and A.mellifera honey.In this study,we designed specifi c primers and probes for A.mellifera and A.cerana respectively based on the MRJP2(major royal jelly protein 2)gene,and developed a TaqMan fl uorescent probe-based real-time PCR assay,and analyzed the specifi city and sensitivity of the method.The results showed that this method was specifi c,and the detection limit was 0.05ng and 0.01ng for A.mellifera honey and A.cerana honey,respectively.About 37%of 54 A.cerana honey samples were authentic.In Conclusion,adulteration of A.cerana honey is very serious,and the detection method has high sensitivity,good specifi city,simple and fast operation,and can provide technical support for the adulteration of A.cerana honey.
