玉米愈伤组织再生体系的研究

摘    要:用玉米胚性愈伤组织作为转化受体时,不同基因型在相同2,4-D浓度的培养基上诱导,其诱导率有较大差别,齐319最高;胚龄对诱导率的影响也较大,10d左右最易诱导,过大或过小均不易诱导胚性愈伤;培养基中2,4-D的浓度对玉米愈伤组织的诱导率有极大影响,不同基因型的最大胚性愈伤组织诱导率所需的2,4-D浓度也不相同,一般为0.5~1.0mg/L。愈伤组织分化时不同基因型所需最适6-BA浓度不同,齐319、515的愈伤组织最适合分化成苗的6-BA浓度为1mg/L,鲁原341愈伤组织最适6-BA浓度为0.5mg/L。

Abstract:Being cultured on the same 2,4-D concentration,calli induction ratios were different among various genotypes,and embryo ages so on.When embryo calli of maize was used to be the receiever in particle-bombardment transformation.It was easy to induce embryo calli about ten days embryo;2,4-D concentration of medium had great efffect on maize calli induction.The great induction ratios of calli on different genotypes need various concentrations of 2,4-D,generally it was from 0.5 to 1.0 mg/L.The properly concentration of 6-BA was different according to various genotypes when calli split up,the suitable concentrations of Q319 and 515 were 1 mg/L,and that of luyuan341 was 0.5 mg/L.
