摘    要:目的]研究不同粗度对灵武长枣(Ziziphus jujuba Mill.)休眠枝水培发芽的影响,并对水培芽无菌外植体的建立进行研究。方法]试验以灵武长枣1年生休眠枝为材料,采用水培催芽的方法在冬季获得外植体。结果]灵武长枣1年生休眠枝茎段基部粗度大于3.5 cm时水培效果较好,抽生枣头较多,生长健壮;以这样的枣头作外植体成活率高达90%以上。外植体消毒灭菌以75%乙醇消毒20 s+0.1%氯化汞消毒15 min+2滴吐温效果良好,污染率为10.0%。结论]该研究获得了灵武长枣休眠枝水培催芽和水培芽消毒处理的最佳方案,可为延长初代培养的取材时限,提高水培芽的消毒处理效果提供科学依据,并为其下一步的试管繁殖、生根等研究提供技术参考。

Germination Status of Annual and Dormancy Branches of Chinese Jujube (Ziziphus jujuba Mill.) with Different Diameter by Water Culture Method and Their Aseptic Explants Establishment in Vitro

Abstract:Objective] The aim was to study annual and dormancy branches of Chinese jujube with different diameter by water culture method,and their aseptic explants establishment.Method] Taking the annual and dormancy branches of Chinese jujube as the material to obtain aseptic explants by water cultured method.Result] The water culture effect was good when the basic diameter of annual and dormancy branches of Chinese jujube was more than 3.5 cm,and the sprouting jujube head was more,healthy and strong.Taking this jujube head as explants,the survival rate increased to over 90%.The effects of surface sterilization was good with 75% ethanol 20 seconds + 0.1% mercuric chloride 15 min + twenn80 2 drops,and the pollution rate was 10.0%.Conclusion] The study obtains the best scheme of accelerating germination of annual and dormancy branches of Chinese jujube by water method and buds sterilizing,which provides a scientific basis for prolonging the time of choosing materials and improving the sterile effects of buds,and provides a technological reference for researches on their tube breeding and striking roots and so on in future.
