大豆CAPS标记快速开发方法的建立与优化

摘    要:采用生物信息学方法,分析大豆SNP位点序列信息,选择合适的内切酶,进行混样PCR和酶切预筛选,建立了大豆CAPS标记的快速开发方法—gspCAPS(Genome Sequence Pool,CAPS)。设计合成了61对引物,在9个大豆品种中对gspCAPS方法进行了评测。结果表明,该方法显著提高CAPS标记开发的效率,传统CAPS方法效率为40.98%(25/61),该方法效率高达86.21%(25/29),并且标记开发的正确性没有降低,维持在100%。本研究所建立的gspCAPS方法效率高、周期短、成本低,对高效开发CAPS标记具有重大的指导意义和广阔的应用前景。

Abstract:In this study,a rapid method,gspCAPS(Genome Sequence Pool,CAPS),was establish-ed to develop soybean CAPS markers.the bioinformatics tools were taken in for analyzing the sequences of soybean SNP sites,and selecting the appropriate restrict enzymes,meanwhile the sample DNA were mixed and the primers were pre-screening by PCR and digested with the restrict enzymes.To evaluate the approach,61 pairs primers were designed and synthesized,and the polymorphism was detected in 9 soybean varieties.The results showed that the approach had remarkably improved the efficiency of the development of CAPS markers,reaching 86.21%(25/29),while the accuracy of the approach wasn't failing,keeping at 100%.It indicated the gspCAPS method was high efficiency,timesaving,and low cost,and was very important to efficient development of the CAPS marker,and had widespread application prospect.
