摘    要: 以棉子油为原料,探索出了一种快速、简便的转基因棉子油的PCR检测方法。该方法能从15 mL棉子油中提取0.4 μg高纯度DNA,用于PCR检测。针对18S rDNA、CaMV35S、NOS、NPTⅡ和Cry1A(c)等基因进行了棉子油中内外源基因PCR检测,分别扩增出不同的目的片段。本研究为油脂类产品的外源基因检测提供了可行方法。

Abstract:A simple and rapid-method which could extract DNA from cottonseed oil had been developed. A yield of 0.4 μg DNA could be obtained from 15 mL oil. PCR amplification of sequences of CaMV35S, NOS, NPTⅡ,Cry1A(c), and an endogenous reference gene 18S rDNA in refined cottonseed oil was performed. Results showed that the sequence-specific fragments were amplified in the samples, demonstrating that the method could be applied for detection of transgene sequences. There are some DNA fragments in refined cottonseed oil, and PCR amplification can detect the foreign DNA.
