基于trnL内含子序列的桑属植物分子系统学初探

摘    要:用PCR产物直接测序法对12份桑种质、1份无花果和1份构树的亮氨酸转运RNA基因(trnL)内含子序列进行了测定。trnL序列长度变异范围为534~561bp,平均核苷酸组成为0.39100(A)、0.27114(T)、0.17679(C)、0.16086(G),平均A+T含量为0.66214,说明该序列富含A/T。利用PHYLIP软件构建其最大简约数(maximum likelihood,DNAML)分子系统发育树,聚类结果表明桑属所有材料聚为一类,进一步证明桑属为单系,为探明桑属植物的亲缘关系提供了新的分子生物学证据。

Abstract:The trnL intron sequences of 12 genus Morus, 1 Broussonetia papyrifera and 1 Ficus carica were amplified by PCR method. Direct sequencing results showed that their length varied from 534 to 561 bp. The average nucleotide composition was 0.391 00(A), 0.271 14(T), 0.176 79(C), and 0.160 86(G), and the mean A+T content was 0.662 14, indicating that the reqion was characterized as AT-rich fragment. Molecular phylogenetic analysis was done by Maximum Likelihood method using PHYLIP software. Clustering results showed that all strains of genus Morus grouped together, indicating that the genus Morus was a monophyletic and phylogenetic relationship among genus Morus was further discussed.
