荧光假单胞菌CZ对TMV的抑制作用及其活性蛋白的分离纯化

摘    要:从烟草根际中分离到一株拮抗烟草花叶病毒(TMV)的生防菌荧光假单胞菌CZ,将其发酵液接种或喷施TMV寄主三生NN烟后进行生物学检测.结果表明:荧光假单胞菌CZ发酵液与TMV混合后接种,其枯斑抑制率为91.5%;接种TMV的前、后24h喷施CZ发酵液,对TMV的抑制率分别为78.9%和30.5%.荧光假单胞菌CZ发酵液经硫酸铵盐析获得活性粗蛋白,经阴离子层析、superdex-G-75凝胶层析和SDS- PAGE检测,分子量为39.4 kD.

Abstract:Pseudomonas fluorescens CZ isolated from soil showed antibiotic activity against tobacco mosaic virus.The results by inoculating necrotic lesions host of Nicotiana tabacum var.Samsum NN showed that the mixture of fermentative liquids of CZ with TMV had an inhibition ratio of 91.5%.The inhibition ratios of inoculated TMV 24 hours after and before spraying fermentative liquid spraying were 78.9% and 30.5% respectively.Active crude protein was extracted by ammonium sulphate salting out from fermentative liquid.Pure active protein was obtained through purification of anion filtration,superdex-G-75 gelatin filtration and SDS-PAGE.The molecular weight of this protein was 39.4 kD.
