灯盏花多倍体植株的诱导培养研究

摘    要:以二倍体灯盏花种子为材料,进行了分别经0.1%的秋水仙素诱导处理12 h、24 h、36 h,接种于培养基培养灯盏花多倍体幼苗的实验研究。结果表明:用0.1%秋水仙素处理灯盏花种子12 h的诱变效果最佳,其所培养幼苗的诱变率为38.9%。通过采用不定芽继代培养的途径对灯盏花幼苗多倍体植株进行了分离与稳定培养,获得了稳定的灯盏花幼苗四倍体植株。与二倍体灯盏花植株相比,四倍体灯盏花植株表现为叶片较厚、较大、叶色较深,气孔也较大,其染色体数目为2n=4x=36。

Abstract:Taking the diploid seeds of diploid Erigeron breviscapus as the materials,the polyploidy induction experiment was conducted by treating the seeds with 0.1 % colchicines for 12h,24h and 36h respectively.The results showed that treating the seeds with 0.1 % colchicines for 12 h had the optimal result of induction rate of 38.9 %.The stable polyploidy plants were screened through adventitious bud culture and plantlet regeneration.Tetraploidy plantlets had leaves thicker and larger than the normal diploid plantlets,moreover,the leaf color was darker and the stomata was larger than the diploid.Cytological observation showed that the chromosome number of variant plantlets is 2n=4x=36,and that of the normal diploid plantlets is 2n=2x=18.
