摘    要:[目的]探索原种黑麦草愈伤组织诱导和植株再生方法,为多年生黑麦草原种的转基因研究奠定基础。[方法]以多年生黑麦草原种成熟种子为外植体,在组织培养条件下,施以不同浓度的激素进行培养,研究不同激素组合对愈伤组织诱导以及植株再生的影响。[结果]MS基本培养基中添加5.0 mg/L 2,4-D时愈伤组织诱导率最高,达64.85%;愈伤组织在MS附加1 mg/L 6-BA和0.5 mg/L IBA的分化培养基中分化率最高,为37%;不定芽在添加0.1 mg/L IBA的生根培养基中生根率为96%。[结论]黑麦草原种愈伤组织再生体系成功建立。

Establishment of Tissue Regeneration System of Lolium multiflorum L.

Abstract:[Objective] The aim was to explore the callus induction and plant regeneration of ryegrass,and lay a foundation for transgenic research of perennial ryegrass.[Method] Mature seeds of ryegrass were used as explants to study the effects of hormones compositions on callus induction,proliferation and plant differentiation.[Result] The result showed that the induction rate achieved the highest of 64.85% on 2,4-D of 5.0 mg/L with 7 g/L Agar.The highest shoot regeneration of 37% was achieved in the medium contained MS medium with 6-BA of 1 mg/L and IBA of 0.5 mg/L.[Conclusion] Ryegrass regeneration system was established by callus induction.
