杏叶片DNA提取和荧光AFLP反应体系的建立

摘    要:以杏嫩叶为材料,利用改进的CTAB法,提取到高质量的杏叶片总DNA,通过优化酶切、连接、预扩增、选择性扩增等试验条件,建立了高效稳定的杏荧光AFLP反应体系;筛选出多态性好的12对引物组合,均获得稳定、清晰的扩增图谱,可进行杏品种鉴定、种质资源的遗传多样性和亲缘关系分析等研究。

Abstract:The high quality DNA required for construction of AFLP analysis system was isolated with an improved method of CTAB from young leaves of apricot.A fluorescent-AFLP reaction system was constructed for genetic analysis of apricot by optimizing DNA restriction-ligase reaction,pre-amplification and selective amplification.The clear and stable amplification patterns could be obtained from 12 pairs primer combinations with good polymorphism,which were selected from 64 pairs AFLP primer combinations.These results ...
