铁皮石斛原球茎固体培养和液体悬浮培养动力学研究

摘    要:为筛选适合铁皮石斛原球茎的最佳培养方式,采用固体培养和液体悬浮培养两种培养方式,并研究液体悬浮培养的营养消耗变化规律,同时考察了液体培养液中pH值和电导率的变化。结果表明:原球茎在固体培养条件下,培养60d后干重达到最大,为13.40g/L,多糖产量最大值为2412.5mg/L;在液体悬浮培养条件下,干重仅需要50d可达到17.64g/L,多糖产量最大值为3081.2mg/L。液体培养在生物量、多糖产量上均优于固体培养;原球茎生长与碳源、氮源和磷的消耗存在着密切关系,培养基电导率变化与原球茎的生物量积累呈线性关系。

Abstract:In order to choose the best culturing method for protocorms of Dendrobium candium,solid culture and liquid suspension culture were used.The variation rules about nutrient consumption were also studied,and the medium pH and conductivity were measured during the culture process. The results showed that:dry weight and the production of polysaccharides reached to it''s maximum 13.40 g/L and 2412.5 mg/L after 60 days solid culture,re sp ectively,but dry weight and the production of polysaccharides reached to it''s maximum 17.64 g/L and 3081.2 mg/L after 50 days suspension culture,respectively. So the liquid suspension culture was superior to solid culture not only in dry weigh but also production of polysaccharides. The growth of protocorms closely correlated with the consumption of sugar,nitrate and phosphate,and the medium conductivity decreased linearly relative to the biomass accumulation.
