| 摘 要： | 通过对冷冻液(1,2—丙二醇浓度和冷冻基础液)以及平衡和脱去防冻剂方法的筛选,研究了山羊卵泡卵母细胞的低温冷冻保存的可能性。结果表明,当冷冻液为含有10％1,2—丙二醇的生理盐水,并以五步法平衡和脱去防冻剂时,冷冻效果最为有效。解冻后,卵泡卵母细胞的回收率和形态正常率分别为96.6％(56/58)和75.8％(44/58),形态正常卵泡卵母细胞的成熟率为16.3％(2/12)。

| Abstract: | Cryopreservation of goat follicular oocytes were studied in the different freezing soloution by the different procedures for addition and removal of cryoprotectant. The results showed that the freezing of goat follicular oocytes can be improved when the 0.9% NaCl solution supplemented with 10% 1,2-propanediol and 20% newborn calf serum was used as cryoprotective solution and cryoprotectant addition and removal was carried out by five-step procedure: The recovery rate and the normal morphological rate of follicular oocytes after thawing were 96.6% (56 / 58) and 76.0% (44 / 58) respectively and the in vitro maturation rate of them was 16.7% (2 / 12). |
