应用同工酶标记区分玉米多胞质系的研究

摘    要:在以往培育成的含Mo17细胞核的班米多胞质系(同核异质系)的基础上,开展了11种多胞质系同工酶生化标记的筛选。通过在不同发育阶段选取不同部位组织样品,进行POD和EST同工酶的电泳分析结果表明,不同发育阶段不同部位组织样品,其同工酶带数与活性是不同的,选取3个发育阶段的不同组织,即授粉后9d籽粒、授粉后34d籽粒乳和抽雄期雄穗作同工酶电泳分析,就可以将11种多种胞质系区分开来。

Abstract:On the basis of previous work, the study of screening marker bands of maize isozyme was carried out. 11 multiplasmic lines used as experimental materials were: (Fli) Mo17 [origin Mo17 inbred line], (su1)Mo17, (sh2)Mo17, (bt1)Mo17, (Pop)Mo17, (wx)Mo17, (Teo)Mo17, (cms T)Mo17 [(T)Mo17], (cms S)Mo17 [(S)Mo17], (cms C)Mo17 [(C)Mo17] and (cms 21A)Mo17 [(21A)Mo17] etc. The different tissues at different development stages were taken for each material. DPAGE system and double vertical gel slabs were used. The experimental results are as folows:(1)POD:A total of 20 isozymic bands. (Fli)Mo17 and (su1)Mo17 possess the POD1 and POD3, while POD7 is absent. (sh2)Mo17 POD15 and POD16. (bt1)Mo17 POD1, POD11. (Pop)Mo17 POD15. (wx)Mo17 does not possess the POD3, 4, 5, 6, 8, 15, 16. (Teo)Mo17 possesses the POD6, 7, 15, but POD6, 8, 10 are absent. (T)Mo17 POD6, 8, 10, 20. (S)Mo17 less POD6, 8, 9, 10, 20. (C)Mo17 possesses the POD5, 6, while POD3, 4, 8, 10, 12, 13, 15, 16 are absent. (21A) Mo17 does not possess the above POD bands.(2) EST:A Total of 18 isozymic bands. (Fli)Mo17 possesses the EST 2, 17, but EST 15 is absent. (su1) Mo17 EST13. (bt1)Mo17 EST6, 12, 14. (Pop)Mo17 possesses the EST12, but EST2, 8, 11, 13, 14 are absent. (wx)Mo17 possesses the EST11, 13, while EST8, 14 are absent. (Teo) Mo17 EST11, 15. (T)Mo17 possesses the EST17, but does not possess EST7, 8, 11, 13. (S)Mo17 possesses the EST2, 17, but does not possess the EST6, 7, 8, 9, 10, 11, 13, 20. (C)Mo17 does not possess the EST7, 8, 11, 13, 17. (21A)Mo17 does not possess the EST7, 8, 11, 13, 17.(3)According to the above data, some specific isozymic bands can be used as biochemical markers to distinguish these multiplasmic lines from each other.
