水稻拟禾本科根结线虫生防细菌的分离鉴定与防治效果评价

摘    要:为获得对水稻拟禾本科根结线虫具有较高杀线虫活性的生防细菌,初筛以马铃薯腐烂茎线虫、复筛以拟禾本科根结线虫2龄幼虫为靶标线虫,采用直接触杀法对土壤分离细菌进行筛选。获得8株具有较高杀线虫活性的菌株,其中D45菌株发酵液的杀虫活性最好。其发酵液上清5倍稀释液处理24、48 h后,马铃薯腐烂茎线虫和拟禾本科根结线虫2龄幼虫的死亡率分别为69.8%、72.1%和60.9%、70.4%。盆栽试验和田间试验表明,在拟禾本科根结线虫侵染前采用D45菌株发酵液进行土壤处理,可显著减少水稻根结数。D45高活性菌株产生的杀线虫物质对蛋白酶稳定,但不耐热和酸。经生理生化测定结合16S rRNA基因序列分析,D45菌株为巨大芽胞杆菌Bacillus megaterium。D45菌株对防治水稻拟禾本科根结线虫具有应用潜力。

Abstract:In order to obtain biocontrol bacteria which can efficiently control Meloidogyne graminicola, soil bacteria were isolated and screened in vitro for their effects on mortality of Ditylenchus destructor and M. graminicola. From primary screening against D. destructor and second screening against second-stage juveniles of M. graminicola, 8 soil bacteria secreting high nematocidal metabolites were isolated. Among them, strain D45 showed the highest activity. After 24 h and 48 h incubation, the supernatant of strain D45 culture diluted five times resulted in a mortality of 69.8% and 72.1% to D. destructor, 60.9% and 70.4% to M. graminicola, respectively. Pot and field experiments indicated that soil drenching with fermentation broth of strain D45 prior to nematode inoculation greatly reduced the numbers of galls. The nematicidal substances of strain D45 were sensitive to protease,while insensitive to acid and heating. Based on physiological and biochemical characters, 16S rRNA gene sequence analysis, D45 was identified to be Bacillus megaterium. This study demonstrates that B. megaterium D45 has good application potential in controlling of M. graminicola.
