芒果炭疽病菌突变体和野生型的生物学特性分析

摘    要:研究了芒果炭疽病菌的突变体和野生型在不同的温度、光照、营养条件下的菌丝生长、分生孢子萌发的情况,研究菌丝体的致死温度和时间,筛选抑制野生型生长的最适潮霉素B和抗除草剂PPT浓度范围。结果表明:糖量多的培养基有利于生长和产孢,菌丝体最适的生长温度为28℃,不同的光照条件下差异不明显,但光照对野生型和突变体的孢子萌发的差异显著。突变体和野生型的菌丝体致死温度为60℃下处理10 min。随着两种添加剂浓度的增加,炭疽菌丝体生长受到抑制,其中潮霉素B在50 mg/kg的浓度下可抑制野生型的生长,抗除草剂PPT在0.02%浓度下抑制生长。

Abstract:In the experiment the mango Colletotrichum gloeosporioides was primarily studied.The effect of different temperature,light and carbon source on the myceliall growth,conidia germination,high thermal death point and time of mycelium was reasearched.And the suitable concentration of Hygromycin B and phosphinothricin(PPT) to wild type was chosen.We concluded that mango Colletotrichum gloeosporioides grew and produced spores better with the growth amounts of sugar.The temperature suitable for its growth was 28 degree Centigrade.Different illumination did not produce obvious difference effect under the controlled condition,but illumination remarkably affected the wild type and spore sprout of mutaion.The lethal temperature for the mutation and wild type was 60 degree Centigrade for 10 minutes.Hygromycin B can suppress wild type growth under concentration of 50 mg/kg and phosphinothricin(PPT) can suppress its growing under 0.02%.The mutant was strengthened with the density but suppressed and grew.
