金柑组培苗对外源精胺的响应

摘    要:为了研究多胺对植物生长发育的调节作用,采用组织培养的方法,以金柑(Fortunella marga-rita)为材料,研究了不同浓度(0mmol/L,0.5mmol/L,1mmol/L,2mmol/L,4mmol/L)的外源精胺(Spm)对金柑组培苗植株生长、根系形态、碳水化合物含量的影响。结果表明,与其他精胺浓度相比较,1mmol/L精胺处理条件下的金柑组培苗的株高(4.87cm)、茎粗(0.122cm)、叶片数(4.2片)、地下部鲜重(0.054 0g)、叶片鲜重(0.087 9g)及植株鲜重(0.174 9g)最高,且其根系的长度(8.77cm)、投影面积(0.86cm2)、表面积(2.70cm2)、平均直径(1.02mm)、体积(0.067cm3)以及其根系和叶片的可溶性糖含量、蔗糖含量均显著高于对照。结论:1mmol/L外源精胺最适合金柑组培苗植株生长和根系的形态发育,这与其提高组织内碳水化合物含量密切相关。

Abstract:Using in vitro Kumquat(Fortunella margarita) as the material,the author studied the effects of different concentrations(0,0.5 mmol/L,1 mmol/L,2 mmol/L and 4 mmol/L) of exogenous spermine(Spm) on plant growth,root morphology,and carbohydrate content to research regulating roles of polyamines on growth and development of plants.The results showed under the conditions of 1 mmol/L Spm,plant height(4.87 cm),stem diameter(0.122 cm),leaf number(4.2 leaves),root fresh weight(0.054 0 g),leaf fresh weight(0.087 9 g) and plant fresh weight(0.174 9 g) of in vitro Kumquat were higher than other treatments.And root length(8.77 cm),root projected area(0.86 cm2),root surface area(2.70 cm2),average diameter(1.02 mm) and root volume(0.067 cm3) of the in vitro Kumquat were significantly higher than the control.Conclusion: 1 mmol/L Spm was the most suitable concentration for growth and root morphology of the in vitro Kumquat,which may be related to increasing carbohydrates of tissue by Spm.
