宁夏枸杞中蛋白质的提取与鉴定

摘    要:采用盐溶法提取宁夏枸杞中蛋白质,用饱和(NH4)2 SO4溶液进行分级盐析、透析、浓缩后用SDS-PAGE分析,测定不同提取液中主要蛋白质的分子量.结果表明:用pH为6.0的PB缓冲液提取,用饱和度为80%的(NH4)2SO4盐析时,枸杞蛋白的提取效果最好;经SDS-PAGE电泳后,初步鉴定出7种主要蛋白质,其中分子量为63 096 Da、51 286 Da、33 884Da的蛋白质提取范围较大.

Abstract:The protein of Lycium barbarum L.in Ningxia were extracted by the salt solvent method,and purified by ammonium sulfate fractionation,dialysis,concentration and the molecular weights of the proteins from different extraction solution were measured by SDS-PAGE analysis.The result showed that the optimum Lycium barbarum L.protein extraction condition was pH 6.0 PB Buffer,80% ammonium sulfate.The SDS-PAGE analysis results showed that 7 kinds of protein were identified,and 3 kinds of protein with molecular weigh...
