雪莲组培过程对潮霉素敏感性的研究

摘    要:以雪莲叶片为外植体,研究了不同浓度的潮霉素对雪莲愈伤组织诱导、生长及分化的影响,结果表明:8.0 mg/L浓度的潮霉素为雪莲叶盘转化筛选的最佳质量浓度,不同浓度(5 ~50 mg/L)潮霉素对MS+ NAA 2.00 mg/L+ 6-BA 1.00mg/L培养基下雪莲愈伤组织的诱导与生长均有明显的抑制作用,随着潮霉素浓度的升高,毒害程度加深,当潮霉素浓度为20 mg/L时表现为生长的愈伤组织块较小、褐化、甚至死亡.同时,低浓度(0.5 ~2.0 mg/L)的潮霉素可以提高雪莲愈伤组织的分化率,其中0.5 mg/L潮霉素的质量浓度的诱导培养基内,雪莲愈伤组织的出芽率平均可达98.33%.

Abstract:To study the effects on induction,growth and differentiation of callus(Saussurea involucrate),the hygromycin and Saussurea involucrate leaves were used as stress and explants,respectively.Cultured on MS+ NAA 2.00 mg/L+6-BA 1.00 mg/L media added different concentration(5 mg/L to 50 mg/L) of hygromycin,the result showed that hygromycin of 8 mg/L was the best concentration for screening calli,there were significantly inhibition on callus induction and growth,and the damagement increased with the concerntration...
