皱皮木瓜超氧化物歧化酶分离纯化研究

摘    要:[目的]探讨皱皮木瓜超氧化物歧化酶(SOD)分离纯化工艺。[方法]以重庆綦江皱皮木瓜为试验材料,采用硫酸铵分级沉淀、Se-vage法除杂蛋白、丙酮再次沉淀脱色除杂和Sephadex G-100凝胶层析,分离纯化皱皮木瓜SOD。采用Folin-酚法测定蛋白质含量,用改良的邻苯三酚自氧化法测定酶活性。[结果]皱皮木瓜SOD在层析过程中得到较好的分离纯化。其纯化倍数是178.72倍,回收率是4.94%,比活力是303.82 U/mg蛋白。SDS-PAGE纯度鉴定结果表明,电泳图为一条谱带,表明分离纯化到达电泳纯的样品。[结论]采用该分离纯化工艺得到比活力较高,达到电泳纯的皱皮木瓜SOD样品。

Abstract:[Objective] This study aimed to discuss the separation and purification technology of Superoxide Dismutase(SOD)from C.spiciosa Nakai.[Method] Using the C.spiciosa Nakai in Chongqing as experiment materials,highly purified samples of SOD were obtained by ammonium sulfate precipitation,Sevage methods,acetone precipitation and Sephadex G-100 gel chromatography.The protein content was measured through Folin-phenol method and the enzyme activity was determined by pyrogallol autoxidation.[Result] SOD of C.spicios...
