苹果实生树嫩芽阶段转变相关蛋白质的SDS-PAGE分析

摘    要:利用聚丙烯酰胺凝胶电泳技术(SDS-PAGE)分析了5株苹果杂种实生树(红玉×金冠)嫩芽中蛋白质含量随节位升高的动态变化。结果表明,随节位升高蛋白质带发生变化,共发现6条阶段转变相关蛋白质带,分子量估算值分别为17.5、19、33、38、51和55 ku。其中55 ku蛋白质表现为定性变化,随着节位的升高,46节出现,之后保持不变,到达125节后又突然消失。17.5、19、33、38和51 ku等5种蛋白质是定量变化。17.5、33、38和51 ku等4种蛋白质在低节位时含量较低,到达一定节位时突然升高,到达较高节位时又突然降低;19 ku蛋白质在5株实生树上的变化趋势不一致,但定量变化的临界点与上述5种蛋白质相同。

Abstract:Dynamic changes of proteins in buds from different nodes of 5 apple hybrid seedlings(Jonathan ×Golden Delicious) were analyzed with SDS-polyacrylamide gel electrophoresis.The result indicated that protein bands changed with nodes along a seedling.6 protein bands presented differences in different ontogenetic phases.The molecular weights of the 6 protein bands were 17.5 ku,19 ku,33 ku,38 ku,51 ku and 55 ku respectively.The 55 ku protein presented a qualitative change,which appeared in the node No.46 and disappeared above the node No.125.Changes of the other 5 proteins 17.5 ku,19 ku,33 ku,38 ku and 51 ku were quantitative.Contents of the 17.5 ku,33 ku,38 ku and 51 ku proteins were low in samples of the lower part of the seedling,when they reached a certain node,the proteins increased to a higher level,and they got to lower level again in higher nodes.Changing tendency of 19 ku in the 5 seedlings was different but critic point of quantitative change was the same with the former 5 proteins.
