摘    要:[目的]苍耳(属)(非中国种)植物为检疫性有害生物,该试验尝试以ITS2序列作为DNA条形码对从国外截获的7种苍耳属植物进行物种区分鉴定研究。[方法]试验应用通用引物对其ITS2基因进行扩增,测序得到7种苍耳属植物的ITS2序列,利用MEGA 5.1软件得到的ITS2序列进行比对和分析,并构建系统树。[结果]共发现变异位点242个,其中单突变位点12个。[结论]ITS2序列能从基因位点层面对苍耳属植物进行区分鉴定。

The Application of ITS2 Sequence as DNA Barcode in Xanthium

Abstract:[Objective] All the species in Xanthium (non-Chinese) are defined as quarantine pests. In this experiment, ITS2 sequence was used as DNA barcode to distinguish and indentify 7 species in Xanthium. [ Method] Universal primer was used to amplify ITS2 genes which were then sequenced. The ITS2 sequences were contrasted and analyzed by MEGA 5.1 software, and the phylogenetic tree was constructed. [ Result ] 242 variable sites and 12 singleton sites were discovered. [ Conclusion] The result shows that the species in Xanthium can be distinguished and indentified by the gene sites of ITS2 sequence.
