麻栎组织培养外植体选择与灭菌方法试验表明：以麻栎播种苗幼苗茎段和大树嫩枝作为外植体进行组培效果较好;播种苗幼苗茎段的最佳灭菌方法是：70%酒精30 s→无菌水洗3次→0.1%升汞2 min→无菌水洗3次;大树嫩枝的最佳灭菌方法是：70%酒精30 s→无菌水洗3次→0.1%升汞5 min→无菌水洗3次;幼苗茎段外植体和大树嫩枝外植体的最佳取材时间是4月,接种后的外植体污染率和死亡率低于其他月份,而芽的萌发率高于其他月份。

Explants selection and sterilization method of tissue culture for Quercus acutissima

Quercus acutissima is widely distributed in China,and it＇s used in many ways such as soil and water conservation and charcoal production.It is important to master the technology of tissue culture for breeding of Quercus acutissima.The results indicated that the seedling stems and the tree twigs were better as the explants of tissue culture than other organ materials.The best sterilization method of the seedling stems explants was soaked by 70% alcohol for 30 seconds → sterile water for 3 times → 0.1% HgCl2 for 2 minutes→sterile water for 3 times.The best sterilization method of the tree twigs explants was soaked by 70% alcohol for 30 seconds → sterile water for 3 times → 0.1% HgCl2 for 5 minutes → sterile water for 3 times.The appropriate season for collecting the explants of seedling stems and tree twigs was in April,because the rates of contamination and mortality of the explants were lower and the germination rates of buds were higher than that in other seasons.
