罗尔斯通氏菌菌株H16新毒素蛋白Reu理化性质的研究

摘    要:为深入研究罗尔斯通氏菌H16新毒素蛋白Reu的理化性质,以初孵菜青虫四龄幼虫为试虫,进行Reu胃毒毒力测定,计算其LD50,检测Reu的热稳定性及其对蛋白酶的敏感性,通过非变性梯度凝胶电泳测定Reu的分子量;SDS-PAGE电泳分析Reu的亚基组成;等电聚焦电泳测定Reu的等电点等。结果表明,Reu对菜青虫四龄幼虫毒力LD50为110.84μg/g;此毒素蛋白经80℃处理10min和蛋白酶K处理后,基本丧失了对菜青虫幼虫的毒杀作用;Reu的分子量为221.3kDa,由64.4kDa和57.5kDa2种亚基组成,等电点为7.08。

Abstract:To study biophysical and biochemical properties of Ralstonia eutropha H16 novel toxin protein Reu,four-age larvae of Pieris rapae was tested.Bioactivity mensuration revealed that Reu had a strong stomach cytotoxicity to Pieris rapae with the LD 50 reaching 110.84μg/g in death.The results obtained from non-denaturing gradient gel electrophoresis showed that Reu is 221.3kDa in molecular weight and comprised of two subunits,64.4kDa and 57.5kDa.Reu was separated into two different protein subunits during SDS-PAGE.Besides,Reu was sensitive to proteinase K treatment and toxicity of Reu was stable within 25-60℃.Its isoelectric point is 7.08.
