基于纳米金合成法评估玉米幼苗耐旱性

摘    要:为检测干旱胁迫前后玉米幼苗的抗氧化活性进而评估玉米幼苗耐旱性,选取‘郑单958’、‘浚单20’、‘登海662’和‘豫玉22’4个玉米品种,采用纳米金合成法对其抗氧化活性进行检测。以透射电镜TEM、X射线光电子能谱XPS表征纳米金,进行条件优化和方法学确证。同时与DPPH法、FRAP法和FC法3种检测抗氧化活性方法的结果进行对比。采用表型鉴定、SOD酶活性和MDA含量测定的方法对玉米幼苗的耐旱性进行检测,并对上述方法检测结果进行相关性分析与主成分分析。结果表明,纳米金合成法检测干旱胁迫前后4个品种玉米幼苗抗氧化活性由强到弱为‘郑单958’、‘浚单20’、‘登海662’、‘豫玉22’,与传统方法和耐旱性检测结果具有一致性。相关性分析和主成分分析结果显示,纳米金合成法与其他方法相关性良好,且在解释总变异中占有较高的表达量,表明用纳米金合成法评估玉米幼苗耐旱性具有良好可行性。

Abstract:To detect the antioxidant activity of maize seedlings before and after drought stress by using gold nanoparticles (GNPs) synthesis method and then evaluate the drought tolerance of maize seedlings, four maize varieties (‘Zhengdan 958’, ‘Xundan 20’, ‘Denghai 662’ and ‘Yuyu 22’) were selected and their antioxidant activity was detected by GNPs synthesis method. GNPs synthesis method was used to test the antioxidant activity, and the conditions were optimized and methodologically confirmed. GNPs were characterized by transmission electron microscope (TEM) and X-ray photoelectron spectroscopy (XPS). At the same time, the results of DPPH, FRAP and FC methods were compared. The methods including phenotypic identification, SOD activity and MDA content determination were used to detect the drought tolerance of maize seedlings. Correlation analysis and principal component analysis were performed on the detection results of the above methods. The results showed that the antioxidant activity of seedlings of the four maize varieties was from strong to weak detected by GNPs synthesis method as: ‘Zhengdan 958’> ‘Xundan 20’> ‘Denghai 662’> ‘Yuyu 22’, which was consistent with the results of traditional methods and drought resistance detection. The results of correlation analysis and principal component analysis showed that the GNPs synthesis method was well correlated with other methods and had a high expression in explaining the total variation, indicating that it is feasible to evaluate the drought resistance of maize seedlings.
