| 摘 要： | 用RT-PCR技术从猪肺泡巨噬细胞中扩增和克隆了猪CD80基因部分片段,经测序鉴定后,用反向PCR技术构建了CD80的缺失竞争cDNA分子。通过竞争PCR方法,建立了CD80标准竞争曲线,并得到其直线回归方程。本方法操作简单,特异性和敏感性高,可用于猪CD80 mRNA水平的定量检测,从而为分析猪的免疫状态提供了一种重要技术手段。

| Abstract: | Porcine CD80 was amplified by RT-PCR from porcine alveolar macrophages and cloned.After sequencing,competitive depletion clones for the cDNA molecules was generated by reverse PCR.Then,the linear regression equation was obtained after construction of the standard competitive curves by quantitative competitive PCR assay.The present study provided an important method to analysis porcine immune status for its simplicity and high specificity and sensitivity during detecting porcine CD80 mRNA level. |
