长蛹龄野桑蚕的AFLP分析

摘    要:采用AFLP分子标记技术对安康学院收集保存的4份长蛹龄和3份普通蛹龄的野桑蚕资源进行遗传多样性分析。结果表明:设计的40对AFLP引物组合中有21对引物组合扩增效果较理想,共获得3806条清晰条带,多态性比率达80.7%,表明野生桑蚕的遗传多样性十分丰富。聚类结果显示:长蛹龄桑蚕之间的遗传距离较大,其中24-1和38-1聚为一类,T-1和10-1聚为另一类。

AFLP Analysis of Wild Silkworm with Long Pupal Instar

Abstract:Using AFLP molecular mark technology, the genetic diversity of four wild silkworm resources with long pupal instar and three wild silkworm resources with normal pupal instar, which collected and preserved by Ankang University, were analyzed. The results showed that among 40 pairs of designed AFLP primer combination, 21 pairs of primer combination had relatively ideal amplification effect, total 3 806 clear bands were obtained, and the polymorphic rate reached as high as 80.7%, thus the genetic diversity of the wild silkworm was very abundant. The clustering results showed that the genetic distance between wild silkworm resources with large pupal instar was relatively long; and among them the 24-1 and the 38-1 was clustered into the same group, and the T-1 and the 10-1 was clustered into another group. Therefore, the genetic relationship between wild silkworm resources with long pupal instar was not close, it should strengthen the conservation and utilization of wild silkworm resources in order to provide germplasm resources for breeding silk worm varieties which suit for production.
