无氯仿提取小麦半籽粒DNA的方法

摘    要:为克服从小麦叶片提取DNA用于PCR受季节的限制以及氯仿不易购买等问题,研究以从叶片中提取的以及用氯仿从小麦籽粒中提取的DNA为对照,用无氯仿方法提取小麦籽粒中的DNA,并用多重PCR进行检测。结果表明,用无氯仿方法提取的DNA浓度比从叶片中提取的和用氯仿提取的DNA浓度要低,而且用无氯仿方法提取的DNA扩增效果稍差,但该问题可通过增加上样量或采用聚丙稀酰胺凝胶电泳等途径来解决。因此,无氯仿小麦籽粒DNA提取方法可在对DNA质量要求不高以及氯仿无法及时得到时使用。

DNA Extraction from Half-Grain Wheat Seed without Chloroform

Abstract:In order to overcome the problems that DNA extraction of wheat leaves for PCR was limited by seasons, and chloroform is difficult to acquire, a method of DNA extraction without chloroform from wheat seeds was carried out in the study, contrasted with extraction method from leaves and the method with chloroform from wheat seeds, and the DNA from the three method was detected with multi-PCR.The results showed that DNA concentration of the trichloromethane free method was lower than that of the two others, and the PCR results reflected that the amplification effect of DNA extracted by trichloromethane free method was a bit weaker than that by the other two methods,but the problem can be addressed through loading more quantity of sample or polyacrylamide gel electrophoresis method. Therefore the trichloromethane free method can be used to with lower DNA quality and under the condition without trichloromethane.
