采用流式细胞仪分析柑橘愈伤组织的细胞周期

摘    要:细胞周期分析可为研究柑橘愈伤组织理化诱变提供依据。试验以暗柳橙、长沙橘、澳洲指橘、纳维来特脐橙等4种培养多年的柑橘愈伤组织为试材,使用流式细胞仪分析其特定时间段的细胞周期,进而找到柑橘愈伤组织理化诱变的最佳时期即分裂最旺盛时期。结果表明,待测4种基因型柑橘愈伤组织在继代2周时处于分裂最旺盛时期,它们的S期时相比例分别为22.4%、21.9%、21.0%和22.9%。同时发现,待测的4种柑橘愈伤组织生长周期长短与再生能力有一定的相关性。极易再生的暗柳橙生长周期最短,易再生的长沙橘、澳洲指橘次之,丧失再生能力的纳维来特脐橙生长周期最长。

Abstract:Cell cycle analysis can provide reference for researching the physical and chemical mutagenesis to citrus calli. Cell cycle phase distribution percentage of 4 citrus calli (Anliucheng sweet orange,Changsha mandarin, Microcitrus, Navelate navel orange) in particular time were measured using the flow cytometry in order to identify the optimum time of their physical and chemical mutagenesis (That was the most vigorous division time). The results showed that the phases of vigorous division and multiplication of the 4 citrus calli were at 2-week-subculture. The S phase highest percentages were 22.4%,21.9%,21.0% and 22.9%,respectively. Moreover, growth cycle length of 4 citrus calli was interrelated with regeneration capacity. Growth cycle length of Anliucheng sweet orange with good regeneration capacity was the shortest, Changsha mandarin and Microcitrus,followed, Navelate navel orange, was the longest.
